Paracetamol And Caffeine Induced Sperm Shape Abnormalities In Mice.

Abstract

Paracetamol [(PCM) acetaminophen] combined with caffeine is widely used as non-prescription analgesic, antipyretic and stimulant, respectively. The indiscriminate utilization of these drugs may pose a serious health problem to the present and future generations. This study evaluated in vivo, the potential genotoxicity of two combinations of PCM and caffeine using the mouse sperm morphology test. Five different doses (mg/kg bw) corresponding to 0.25x, 0.5x, 1.0x, 1.5x and 2x of the maximum daily human therapeutic doses (HTD) of each of the 2 drugs were considered. Mice were given 0.5ml of each dose per day for five consecutive days by intraperitoneal injection (IP). Each dose group comprised five mice, and a 5-week exposure period was utilized. Normal saline and cyclophosphamide (20mg/kg bw) were used as the negative and positive controls, respectively. Data obtained show that the test drugs induced a dose-dependent, statistically significant increase (P
